PART ONE:EXPRESSION OF PROTEINS AND GENES INVOLVED IN NEUROTRANSMITTER RELEASE AND SYNAPTIC VESICLE RECYCLING IN PHT RESISTANT KINDLING RAT MODELSObjective To study the changes of proteins and genes involved in neurotransmitters release and synaptic vesicle recycling in the brains of PHT resistant kindling rat models, demonstrating synapse function changes in intractable Epilepsy.Methods built PHT resistant and PHT-effective epileptic rat models, studied their brain cDNA microarray, identified the differentially expressed genes involved in neurotransmitters release and synaptic vesicle recycling , then used western blotting and immunohistochemistry to evaluate the differentially expressed proteins involved in synaptic vesicle recycling.Results As compared with PHT-effective epileptic rat models, PHT resistant group appeared increased mRNA expressions of clathrin, synaptotagmin I ,VAP33,syntaxin 7 ,dynein,γ- SNAP,et al,who were involved in neurotransmitter release and synaptic vesicle recycling, decreased mRNA expressions of HSC70 pseudogene, caldesmon, gamma-actin, and synaptogyrin3; appeared significant increase in clathrin...
