Objective: Ubiquitin proteasome system (UPS) existed in all eukaryotic cells, is playing a central role in selective degradation of intracellular proteins, through which, up to 80%-90% of all intracellular proteins are degraded by UPS. UPS not only modulate the levels of the proteins, but also is concerned with the control of inflammatory processes, cell cycle regulation and gene expression. A series of pathological changes should occure when the function is abnormal,so it is very important to maintain normal function of UPS. As well known, cells apoptosis and inflammatory processes are important mechanism of myocyte ischemia/reperfusion injury and nucleus factor kappa B (NF-κB) plays a critical role in the development of inflammation and cell apoptosis. Because the activation of NF-κB is increased during ischemia/reperfusion, proteasome inhibitor may attenuate ischemia/reperfusion injury and cell apoptosis through inhibiting activation of NF-κB.
