Objective:As a familiar disease, Apoplexy has taken place frequently and the ratio of death and lameness are both very high. Moreover, the cerebral ischemia accounts for 75 percent of Apoplexy, it also has a tendency for increasing year after year. So it is an urgent problem for us to prevent and cure the cerebral ischemia or to decrease the ratio of incident and lameness. From the foregone clinical and experimental research, we have found that electroacupuncture (EA) can improve the clinical symptoms and diversified bio&chem indexes. So this research is to observe the effect of EA on the expression of TNF-α mRNA, NF-κB and P53 in the model rats' cerebral ischemia area and the content of TNF-α and NO of cortex and also the neuron apoptosis in cortex by EA on model rats' shuigou and neiguan (left) point, and study the protective effect and it's mechanism of EA on the cerebral ischemia. Consequently we investigate the mechanism of treating by acupuncture and moxibustion, and after the theory basis for acupuncture and moxibustion treating ischemic cerebrovascular disease in clinic. Method:Healthy and grown SD rats are randomly and evenly assigned to normal group, sham-operation group, model 12h group, model 24h group, model 72h group, EA treatment 12h group, EA treatment 24h group, EA treatment 72h group, Cerebral ischemia reperfusion model is established by occlusion of the middle cerebral artery for 30min and then removal of the occlusion for 12,24 and 72h respectively, then EA shuigou and neiguan left points of experimental rats.We observe the morphological change of experimental rats by HE staining, detect the neuronal apoptosis in cortex by the terminal deoxynucleotidyl transferase mediated dUTP-biotin nickend labeling (TUNEL), observe the change of TNF-α mRNA in cortex of ischemia area by RT-PCR, measure the change of TNF-α and NO in the cortex by radioimmunoassy (RIA),detect nuclearfactor NF-k B(NF- k B) and P53 protein in the ischemia cortex by immunohistochemistry.
