BackgroundChronic myelogenous leukemia (CML) is a clonal malignant disease of hematopoietic stem cell , and it had CML stem cell with CD34 antigen positive accompanied by BCR/ABL fusion gene positive.The study shown that this kind of cells had special biological characteristic,and play an important role in CML onset and development, and it would effect the clinical therapy result and the prognosis of CML patients.Besides allo-HSCT, STI571 is well known to be the best drug to prevent CML progress from chronic phase to blastic crisis. But the disease will relapse in patients whose therapy was discontinued even after CCR. By the help of Q-RT PCR, we can find a few CML cells still exsist in these patients. And in previous study, we also find in some patients with CCR after STI571 therapy, the bone morrow MNC was BCR/ABL negative by FISH analysis, but these patients would relapse after auto-HSCT. We studied CD34~+ cells purified from these patients' bone marrow and also found the expression of BCR/ABL fusion gene in these cells. These results demonstrate that STI571 can't eliminate BCR/ABL~+-CD34~+cells, and these cells will become a source of CML recurrence.The biological characteristics of BCR/ABL~+-CD34~+ cells and the way to...
