Radix Bupleuri (Chai-Hu) is one of the commonly used Traditional Chinese Medicines (TCM). 24 samples of Radix Bupleuri including 10 species were collected from different habitat and were studied on the fingerprint of the raw materials. Firstly, HPLC method was applied to analyze saikosaponins and flavonoid of all samples and 38 constituents were eluted. The chemical data was obtained and studied by chemical pattern recognition methods. All samples were identified with hierarchical clustering analysis and divided into three grades: the superior, the general and the inferior. The superior samples were selected to develop the common recognition Pattern. The similarity analysis method for quality assessment of Radix Bupleuri samples was built by cosine, correlation and Euclidean. Secondly, CGC method was applied to analyze essential oils of all samples and 40 compounds were found. The chemical data was obtained and studied by chemical pattern recognition methods. All samples were identified with hierarchical clustering analysis and divided into six grades. The superior samples were selected to develop the common recognition Pattern and the similarity analysis method for quality assessment of Radix Bupleuri samples was built by cosine, correlation and Euclidean.The developed HPLC and CGC chromatographic fingerprints were different for evaluating the quality of Radix Bupleuri. Therefore, it was necessary that the quality of Radix Bupleuri should be evaluated by the two fingerprints. The developed method was reasonable and feasible to describe and evaluate the quality of unknown samples based on the chemical data. The methodology might well be used to assess the quality of other herbs.The chemical compositions of essential oils from ten Bupleurum species have been studied by GC-MS method, Kovats retention indices method etc. Fifty constituents of the essential oils were identified.
