Objective: To analyze the incidence and profile Of mutations in epidermal growth factor receptor(EGFR) in Chinese patients with the lung cancer. Methods: A total of 80 cases of lung cancer tissue was collected. 50 cases the normal lung samples were also available.The tissue DNA was abstracted and the EGFR gene in exons 19 to 21 was subjected for PCR amplification and direct sequencing. Result: Mutations were found in 36 cases of 80 samples, with the incidence of 45.0%. There were 6 types of EGFR mutation in exon 19, including delE746-750 (1), delE746-A750 (2), delL747-E749insP, del L747-A750insP del L747-S752 , delE746-S752insV, resulting in the deletion of the codon 746 to 752. There were 2 types of EGFRmutation in exon 20 (1789F-M793F was detected in neoplasitc lung tissues,V785F in normal lung tissues ).The mutation in exon 21 belonged to the single missense substitution in the codon 858 . EGFR mutation was found significance as to gender, age and lymphous metastasis, but its mutation was found significance as to cell types and the status of smoking.
