Objective: To explore the regulated mechanism of apoptosis in the pathogenisis and development of the craniopharyngioma ,and to find the relationship of the apoptosis mechanism and the prognosis. To discuss the genetic changes of the pathogenisis and development of the craniopharyngioma. To investigate the clinical factors to affect the prognosis of the craniopharyngioma. Methods: (1) Our research includes three parts.(1)We use the opticmicroscope electronmicroscope and TUNEL technique to observe the morphologic character of 50 cases of the craniopharyngioma ,and quantitative analysis the number of apoptosis cells in the craniopharyngioma with apoptosis index. With the immunohistochemistry method ,We detect the expression of the Bcl-2 and Bax genes in the tumor cells of 50 cases of the craniopharyngioma, and calculate the ratio of Bcl-2/Bax. (2)We design the plan of case-control, and detect the expression of the β-catenin gene in the tumor cells in the craniopharyngioma group and the normal nerve cells in control group. With the techniques of PCR DHPLC and DNA sequencing,We screen and analysis the mutation of the extron 3 of CTNBB1 of tumor tissue of craniopharyngioma of different pathotypes and the normal nervous tissue...
