Objectives: Nano material-mediated gene therapy has become a novel strategy for cancer treatment. Nanoparticles as a drug delivery system will play a big role in gene therapy. Currently gene therapy has been a promising clinical approach for treatments of cancer and genetic diseases. The key step of gene therapy is how to accomplish the gene transfer, which is to introduce the exogenous gene into the cells. Application and development of gene therapy have been restricted for two reasons. One is the potential safety problems caused by toxicity and immunogenity of the classic viral or non-viral delivery system; another is the low transduction efficiency. Nanoparticle gene transfer technology utilizes the nanoparticle as vehicle, which is to encapsulate or attach DNA or RNA gene therapeutic molecules. Because of their small size, large surface area to volume ratio(that is great accessible surface area), functional groups on nanoparticles and selective binding affinity enhance;the time required to achieve binding equilibrium shortens greatly.
