Objective to seek for a method of anti-oxidizing injury by the combination of pulmonary surfactant ( PS) and superoxide dismutase (SOD), as well as a way to enchance the transport of PS-SOD liposome into lung tissue, so as to lessen the injury of lung tissue caused by CPB,hyperxia and NO. The result may be helpful to future clinical work. Methods 1) SOD is labeled by FITC through Clark's method. Its labeling rate is detected through ultraviolet spectrophotometer. The activity of SOD is detected with SOD kit just before and after the labeling. 2) PS- SOD liposome is combined through rotation-evaporation method. Electronic microscope is used to measure the diameter of liposome. Surface tensometor is used to measure surface tension of PS-SOD liposome.The next step is to detect the combined rate of the liposome and the activity of SOD in the liposome. 3) The liposome is instilled into rats lung through trachea. Then the rats are killed by decapitation at 2h and 24h after the instilling. The fluorescence intensity of rats lung is analysed with fluorescence microscope. After the Lavaging and homogenating of rats lung, the biochemical activity of SOD in lavage and homogenate are analysed with SOD kit. All results are analysed with SPSS 10.0 software. Results 1) SOD can be labeled successfully by FITC through Clark's method. Its activity is not affected. The F/P rate is 2.76. 2) PS-SOD liposome pan be synthesised through rotation-evaporation method with the dimeter of 0.463?.223um. Surface tension of PS-SOD liposome is not affacted.The entrapment rate is 40% and the activity of SOD is not changed significantly. 3) By analysing the quantity and quality of flourance in lung microtome, the uptake of SOD into ratslung is much higher in liposome group than in other groups. A similar result can be got by analysing the biochemical activity of SOD in lavage and homogenate. Conclusion SOD can be labeled and PS-SOD liposome can be synthesised successfully.
