Recently, with the major development of light scattering theory and technology, large number of important light scattering analytical technique have been emerged, they have been widely applied to the research of physical chemistry, polymer chemistry, colloidal science, and life science, and played an important role in the study of property, bio-assemblies and aggregation of bio-molecules. The resonance light scattering (RLS) technique, which was by Pasternalk let, not only meets high sensitivity but also offer the additional benefits of simplicity and versatility. Nowadays it has become a new spectral analysis technique and has been found wide applications in the analysis of proteins, nucleic acid, drugs, carbohydrates, metallic ions, surfactants, biomolecules, and nanoparticles. However, light scattering has suffered from drawbacks such as interference caused by other particle in solution and the deficiency of analytic selectivity, which causes some difficulties to the application of it. In this study, started with overcoming the limitation of light scattering technique and guided by the theory and technique of RLS, we try to combine the advantages of the high sensitivity and easy operation of RLS technique with the specificity of immunoreactions, and construct a novel homogeneous immunoassay based on the measurements of RLS signals with a common spectrofluorometer. On the basis of it, the study further applies the immunoreaction into solid support, and develop a new immuneosensor system based on surface enhanced light scattering (SELS) measurements, which not only develop the practical fields of the RLS technique, but enrich the types of immunosensor.The main contents and some conclusions of the thesis are as follows:1. Applications of resonance light scattering technique in immunoassay. Experimentsindicate that the pure antigen and antibody has very weak RLS signals under the lowconcentration of antigen and antibody, but the strong RLS signals could be observedobviously after antigen and antibody take the immune precipitation reactions.
