Immunoliposomes are liposomes which have antibody attached to the nembrane surface. Linking the antibody to the vesicles enables the liposomes to be directed to a specific target, meanwhile the liposome preserves its own characteristics. There could be about 1,000 to 10,000 molecules of drug encapsulated in one immunoliposome without any change of chmical structure of the drugs. This allows a wide range of water soluble or lipid soluble molecules of verying sizes, small ions to macromolecules, to be entrapped. Therefore, immunoliposomes are an effective tool for targeted drug delivery for almost all kinds of drugs and are gaining attentions in immunodiagnosis and treatment of cancer. In the present study, the anti-gastric cancer monoclonal antibody MGb2 was conjugated with liposomes encapsulating antinoeplastic agents: doxorubicin, palmitoyl daunorubicin, epirubicin, mitomycin C, methotrexate, cis-platin, actinomycin D, bleomycin, vincristine and two kinds of boron-10 compounds; thus, 9 kinds of anti-gastric cancer immunoliposomes and 2 kinds of boronated immunoliposomes were prepared. The characteristics and in vitro cytotoxicities were compared and their in vivo biodistribution and and antitumor effects upon human gastric cancer in different nude nice xenograft models with different routes of administration were investigated.Preparation of immunoliposomes: Liposomes were prepared with improved reverse-phase evaporation method to get reverse-phase evaporation vesicles (REV) or with sonication method to get small unilamellar vesicles (SUV) encapsulating 9 kinds of antineoplastic agents and 2 kinds of boroncompounds seperately during the preparation procedure.
