INTRODUCTION In previous study, we have shown that Ehrlich ascites tumor cell (ET) treated with chemicals appeared promising to act as tumor vaccines with safety and effectiveness. Immunizing mice by injection of modified autologous tumor cells vaccines could elicit protective immune response.AIM In order to elucidate this important phenomenon, we studied thepreparatory characteristic of the tumor vaccines and the mechanisms of the immune response. Here an attempt was made to observe the characteristics of the vaccines in the respects of cell cycle and ultrastructure study and examine the cellular and humoral immune response in the immunized BALB/c mice. METHOD Cell cycle analysis and the ultrastructure study of the vaccines were made by fluorescence-activated cell sorter (FACS) method and electron microscope respectively. Para-vaccine sections were stained by terminal deoxynucleotidyl transferase mediated deoxyundine triphosphate biotin nick end labelling (TUNEL) to identify apoptotic cells. DTH response was used as an indicator of cellular immune response in vivo. Proliferative capacity of splenocytes was examined by ~3H-incorporation, and phenotype and release of IFN-gamma in an ELISA assay of effector cells after multiple restimulations of splenocytes in the presence of modified tumor cells (MLTC) were also measured in vitro.
