The occurrence of tumor is related to a series of changes of gene,including the activition of oncogene and the inactivition of anti-oncogene.They all can lead to unlimited proliferation of cells and result in tumor.But normal mammalian cells have a suit of elaborate regulative mechanism which balance and restrain the uncontrolled cell proliferation .One of them is apotosis.Another is the progressive loss of telomere DNA with each cell division. Telomere is necessary for the stabilization of chromosomes. The continuous loss of chromosome telomere lead to unstable of chromosome, and result in senescence or cell death. Maintenance of telomere length is critical to the efficient proliferation of eukaryotic cells. Telomerase is a ribonucleoprotein complex that is thought to add telomeric repeats to the ends of chromosomes during the replicative phase of the cell cycle,which function to overcome cellular senescence and are linked to unlimited cell proliferation and become immortal cells. The obtained immortalization of cell is the key step of cell canceration. Telomerase is active in about 90% tumor tissues , immortalized cell lines and in human germ cells. In contrast, most mature somatic cells and tissues contain low or undetectable telomerase activity. Thus,there's a close relationship between telomerase activity and the cccurence of tumor .Telomerase should be possible to act as a specific tumor marker and an ideal cancer target.Due to shortened telomere and telomerase expression of a majority of malignancy tissues ,many people believed that telomerase was reactivated in the process of canceration. Telomerase activity was regulated. At present ,the study on the regulation of telomerase is one of the hotspots. To further study on telomerase regulation may offer the new idea for designing antitumor drugs. Widespread hematopoitic system malignant tumors display shortened telomere and express telomerase. The level of telomerase activity is different in different hematopoitic system malignant tumors and highest in acute leukemia. In the different progressing phase of leukemia ,the telomerase is particularly activited and the level of telomerase activity consistent with the progressingof leukemia. The patient of leukemia with high level telomerase activity has bad prognosis and survival rate . It is a potential method to treat leukemia by understanding the telomerase activity ,telomere state and the regulation of telomerase activity ,and regulating telomerase activity to treat leukemia.
