The researches and applications on antibody molecules are always the hotspots of immunology research, even all biology researches. Antibodies are attractive therapeutic and diagnostic reagents, and account for a majority of all bioproducts. Over the last century animal antiserum (polyclonal antibodies) was used, with the successful development of hybridoma technology, numerous monoclonal antibodies have been generated for used in diagnosis and therapy of human diseases over the past 20 years. Subsequently, the use of recombinant DNA technology permitted the genetic manipulation of antibody molecules. The third generation of antibody - genetic engineering antibodies initiated a new era for science. Hepatitis A is one of significant public health problems. To date, there are several kinds of HAV vaccines to prevent it, and immunoglobulins were always used to passive immunoprophylaxis for contact with hepatitis A patients without administering vaccine or travel to endemic areas. Although immune serum globulins have high anti-HAV titers and provides effective protection, they may be contaminated by other causative agents in blood. Human genetic engineering antibodies may be the preferred pharmaceutics. Phage antibodies against HAV were screened from a human antibody library by using antibody-capturing HAV antigen, and then the expression vectors for Fab and whole human IgG molecules were constructed, and anti-HAV Fab and whole IgG were expressed in different expression systems.1.
