Objectives: The incidence and mortality of lung cancer increased fastly in recent years. Lung cancer has become one of the major malignant diseases that threaten the health of Chinese people. It is imperative to find some new methods to treat it. After operation, a large portion of patients still died from cancer reoccurance and/or metastasis because their immunity has been victimized. Recently, immunotherapy has become a promising candidate to improve 5-year survivals.Adoptive cellular immunotherapy (ACI) is currently one of the major methods in immunotherapy. Cells that can be used in ACI include: Lymphokine-activated killer cell (LAK), Tumor-infiltration lymphocytes (TIL), and Cytotoxicity T lymphocytes (CTL), etc. LAK and TIL have been used in clinic. However, LAK is not so specific and its rule needs large quantity of IL-2. It was used limitly because the side effects of IL-2. TIL is mainly composed with CD3+T cell. Its cytotoxic activity to autologous tumor cells is very few. The effect of ACI depends on the amount of effector cells and frequency of inoculation. Hypothesisly, increase the amount of effect cells and frequency of inoculation might improve the effects of the treatment. In present research, we tried to find a method to get much more effector cells. These cells should have great cytotoxic activity to the tumor and no harmful to normal cells. The amount of effector cells harvested should be sufficient for clinical use.Material and Methods: Part one: The culture and identification of tumor drainage lympn nodes (TDLN) cells. Two to four lymph nodes without metastasis from lung cancer drainage region were removed during operation. The single cell suspension of TDLN cell was obtained and suspended in 30ml of RPMI 1640 medium containing IL-2 175 U/ml (I-TDLN group) or IL-2 175...
