AimsOwing to the impaired immune function of lymph nodes,it is easy for lymph node to act as routes for tumor cells toward distant organ metastasis.Many studies had showed that the decline of the immune cells and changes in the proportion of T cell subsets would result in the immune suppression.In this study,we compared the number of T lymphocyte subsets from lymph nodes in cancer patients without lymph node metastasis with those from the tumor draining lymph nodes in cancer patients with lymph node metastasis,then analyzed their difference of immune function.In addition,we designed the experiment to explore the effect of interleukin-2 on the activation of the T lymphocyte subsets,discussed the possibility of interleukin-2 as a drug to improve the immune function of lymph nodes in cancer patients.Methods1.Breast cancer and thyroid cancer patients in May 1,2016 to December 31,2016 of Jinling hospital were included in this study.According to the postoperative pathology results they were divided into the control group and the observation group.The control group(13 cases,7 cases of breast cancer patients,6 cases of thyroid cancer patients)patients were diagnosed with lymph node metastasis;the observation group(13 cases,6 cases of breast cancer patients,7 cases of thyroid cancer patients)patients were diagnosed without lymph node metastasis.We obtained the lymph nodes of cancer patients during the surgery,then measured the number of CD3+CD4+T cells and CD3+ CD8+ T cells in lymph nodes using the immunohistochemical staining technique,measured the concentration of interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells using the enzyme linked immunosorbent assay.T test was used to compare the differences of CD3+CD4+T cells,CD3+CD8+ T cells,interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells in two groups,then we analyzed the differences between two groups.2.Breast cancer and thyroid cancer patients in May 1,2016 to December 31,2016 of Jinling hospital were included in this study.According to the postoperative pathology results they were divided into the experimental group A and the experimental group B.The experimental group A(11 cases,6 cases of breast cancer patients,5 cases of thyroid cancer patients)patients diagnosed with lymph node metastasis were treated with interleukin-2 before operation.The experimental group B(11 cases,4 cases of breast cancer patients,7 cases of thyroid cancer patients)patients diagnosed without lymph node metastasis were also treated with interleukin-2 before operation.We obtained the lymph nodes of cancer patients during the surgery,then measured the number of CD3+CD4+T cells and CD3+CD8+T cells in lymph nodes using the immunohistochemical staining technique,measured the concentration of interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells using the enzyme linked immunosorbent assay.T test was used to compare the differences of CD3+CD4+T cells,CD3+CD8+T cells,interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells in each groups.We analyzed the causes of the differences,then discussed the possibility of interleukin-2 as a drug to improve the immune function of lymph node in patients with cancer.Results1.The number of CD3+CD4+T cells,CD3+CD8+T cells,the concentration of interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells in the observation group were higher than those in the control group,and the differences were statistically significant(P<0.05).2.The number of CD3+CD4+T cells,CD3+CD8+T cells,the concentration of interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells in the experimental group A were higher than those in the control group,and the differences were statistically significant(P<0.05);The number of CD3+CD4+T cells,CD3+CD8+T cells,the concentration of interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells in the experimental group B were higher than those in the observation group,and the differences were statistically significant(P<0.05).In addition,the number of CD3+CD4+T cells,CD3+CD8+T cells,the concentration of interleukin-2 and interleukin-4 secreted by CD3+CD4+T cells in the experimental group B were higher than those in the experimental group A,and the differences were statistically significant(P<0.05);ConclusionsIn summary,the findings from this study showed that the lymph nodes of patients without lymph node metastasis had more normal immune function compared with those of cancer patients with lymph node metastasis.In addition,the application of interleukin-2 could significantly improve the T cell immune function in the tumor draining lymph nodes.Interleukin-2 might be a potential drug to recover the impaired immune function of tumor draining lymph nodes and used in the treatment of cancer.
