Effects Of Qingdu Tablet And Antisense Oligodeoxynecleotide Targeting HTERT MRNA On Acute Leukemia

In order to obtain new method to treat acute leukemia(AL), we studied the effects of Qingdu tablet and antisense oligodeoxynecleotides(ASODN) targeting human telomerase reverse transcriptase mRNA(hTERT mRNA) on acute leukemia.1 Studying of documents1.1 Objective: to offer theory references for this study.1.2 Method: By reviewing documents, the relationship between telomerase, hTERT, Antisense oligodeoxynecleotides and AL was analyzed and summarized, we also analyzed and summarized the mechanism of action of traditional Chinese medicine for curing AL.1.3 Result:1.3.1 Activation of telomerase is a important part in form of malignant tumor. hTERT is a determinative fator regulating the activation of telomerase. Telomerase and hTERT mRNA express high activation in AL and cell line of AL. It is a new method to anti-acute leukemia by restraining the activation of telomerase.1. 3. 2 Antisense oligodeoxynecleotide can restrain, seal and destroy target gene by combining definite sequence of gene, then to disturb the transmit of genetic information from nucleic acid to protein. ASODN targeting hTERT mRNA can restrain the activation of telomerase.1. 3. 3 TCM can treat AL by inducing apoptosis, inducing cell differentiation, deteriorating drug resistance, mediating immunological regulation and con-troling gene expression.1.4 Conclusion: In abstracto, TCM is used combining ASODN targeting hTERT mRNA can obtain synergistic therapeutic action because both TCM and ASODN cancontroll gene expression.2 Clinical research2.1 Segment 1: Study of the relationship between type of syndrome of TCM and expression of hTERT mRNA.2.1. 1 Objective: To explore the nature of TCM about the expression of hTERT mRNA, and to offer theory reference for finding suitable TCM to interfere the expression hTERT mRNA.2.1. 2 Method: The expressions of hTERT mRNA of peripheral blood monouclear cells(PBMC) of 29 AL patients were measured by half quantitation RT-PCR. 20 matched control subjects were investigated in this study.2.1.3 Result: The expressions of hTERT mRNA in AL patients were significantly higher than that in the control group(P<0. 01). The expressions of hTERT mRNA in AL was in following sequence:stagnant blood-sputum type Qingdu added ASODN group> ASODN group> Qingdu group. Time dependence was shown in each group.3. 3. 2 Effect of Qingdu tablet and hTERT ASODN on apoptotic of HL-603. 3. 2.1 Morphology of apoptotic:Treated by drug, HL-60 cell have the change of apoptotic including cell body diminution, cell membrane complete, kytoplasm vacuolus, chromatic agglutination, karyopycnosis, and so on.But there is no change in negative control group and blank group.3. 3. 2. 2 Annexin V-FITC/PI double label method with FCMThere was no significant difference in apoptotic rate between negative control group and blank group. There was very significant difference in apoptotic rate between Qingdu added ASODN group and Qingdu group or between Qingdu added ASODN group and ASODN group. The apoptotic rate was in following sequence:Positive control group> Qingdu added ASODN group) ASODN group> Qingdu group>negative control group. Time dependence was shown in each group.3. 3. 3 Effect of Qingdu tablet and hTERT ASODN on expression of hTERT mRNA of HL-60There was no significant difference in expression of hTERT mRNA between negative control group and blank group. There was very significant difference between the group added drug and negative group or blank group. The expression of hTERT mRNA was in following sequence: Qingdu added ASODN group> ASODN group> Qingdu group>Positive control group>negative control group. There was significant difference between Qingdu added ASODN group and other group.3.4 Conclusion: There is synergistic effect in cytostasis, inducing apoptotic, decreasing expression of hTERT mRNA when combined Qingdu tablet with ASODN, and time dependence was shown in each group. Combining TCM with ASODN may be a new method to treat AL.
