Objective:To observe the influence of electroacupuncture at Shu-points of the five Zang-organ for the classical animal model—acute allergy neuritis domestic rabbit's peripheral nerve pathology morphology ultrastructure and the neuroelectricity physiology function as well as to the sciatic nerve organizes Helper T cell subsets TNF- α, IL-4, IL-10 , IL-12mRNA and the costimulatory molecules CD28 , CTLA-4 , B7-1 , B7-2protein expression level;to discuss the function of Helper T cell subsets and the costimulatory molecules in the mechanism of EAN;thus, to promulgate the mechanism of the treatment of EAN by electroacupuncture at Shu-points of the five Zang-organ and rest on the theory of " governs flaccidity alone to take Shu-points of the urinary bladder five Zang-organs."Method: We intensively mixed the cow P2 3ml and the isometric CFA, the immunity domestic rabbit prepared the EAN animal model. This experiment totally used 80 domestic rabbits which divided into four groups. 20 normal healthy domestic rabbits are the blank group, the blank group replaced the cow P2 and isometric CFA by the 3ml physiological saline carry on the immunity to take the comparison, moreover took 60 EAN domestic rabbit which the mold succeeded to randomly divide into the modelgroups, the electroacupuncture group, the immunity globulin group, each group had 20 cases. The electroacupuncture group gave electroacupuncture at Shu-points of the five Zang-organ and the immunity globulin group gave the rabbit immunity globulin 80mg/ (kg*d) intravenous injection.In the 7th days and the 14th days we separately examined each group of experimental animal sciatic nerve movement conduction velocity and the F wave appearance frequency, and selected the material from the experimental animal, and used the electricity to observe the change of the peripheral nerve pathology morphology ultrastructure;using RT-PCR to examine the expression level of Helper T cell subsets TNF-a ? IL-4> IL-10> IL-12mRNA;using ELSA to examine the protein expression level of sciatic nerve the costimulatory molecules CD28> CTLA^ B7-K B7-2.Result1. We observed the model groups domestic rabbit sciatic nerve mainly pathology performance is myelin sheath swelling and myelinoclasis, with the chondriosome tumescent, the schwann cell nucleus pyknosis, the cytoplasmic matrix cavitation, the cell organ swelling. After the treatment, the electroacupuncture group and the immunity globulin group's myelinoclasis status had a distinct improvement, and the readability of myelin sheath lamina had a enhancement, with the thick nerve fiber structure basically normal, with the chondriosome individual tumescent, the schwann cell organaswelling decrease, cellular form basically restoration, newly tiny marrow nerve fiber production.2. The movement conduction velocity of EAN domestic rabbit sciatic nerve obviously reduced. The movement conduction velocity of sciatic nerve in the electroacupuncture group was obviously better than in the model groups (P<0.01), with no significant differences as compared with those the immunity globulin group (P>0.05) in the 14th day;the F wave abnormal cases of EAN domestic rabbit sciatic nerve obviously increased. The F wave abnormal cases in the electroacupuncture group was obviously reduced (P<0.05), with no significant differences as compared with those the immunity globulin group (P>0.05) in the 14th day.3 . In the model groups, the mRNA expression of TNF- a > IL-12 obviously advanced, while after the treatment it dropped obviously. There is no significant differences between the electroacupuncture group and the immunity globulin group (P>0.05) in the 14th day. While in the model groups, the mRNA expression of IL-4^ IL-10 obviously increased after the treatment, with no significant differences as compared with those the immunity globulin group (P>0.05). There was no significant differences between the electroacupuncture group and the immunity globulin group (P>0.05) in the 14th day.4. Comparing the model group with the blank group, the expression level of CD28, B7-1 started to elevate in the initialperiod, after the treatment it obviously reduced (P<0.05). There was no significant differences between the electroacupuncture group and the immunity globulin group (P>0.05) in the 14th day. Comparing the model group with the blank group, the expression level of CTLA-4, B7-2 was lowest in the acute stage (P<0.01), after the treatment it obviously reduced (P<0.05). There was no significant differences between the electroacupuncture group and the immunity globulin group (P>0.05) in the 14th day.Conclusion1 . The myelin sheath swelling and myelinoclasis of EAN domestic rabbit sciatic nerve got improved after electroacupuncture at Shu-points of the urinary bladder five Zang-organs, while the chondriosome and the schwann cell organ swelling reduced, the cellular form restored basically.2. The movement conduction velocity of EAN domestic rabbit sciatic nerve obviously elevated after electroacupuncture at Shu-points of the urinary bladder five Zang-organs, while the frequency of the sciatic nerve F wave reduced.3. Helper T cell subsets existed unbalance in EAN. The Thl cell got the advantage at disease recovery period, while the Th2 cell at the peak. Electroacupuncture at Shu-points of the urinary bladder five Zang-organs could antagonist the unbalance of Th cell subsets and caused the mRNA expression of Thl cell to reduce obviously while the mRNA expression of Th2 cell to elevate.4. Costimulatory molecules CD28 and B7-1 which had the immunity raise function, CTLA-4 and B7-2 which had the decline function participated the morbidity of EAN. Electroacupuncture at Shu-points of the urinary bladder five Zang-organs could reduce the expression level of the costimulatory molecules CD28 and B7-1, while the mRNA expression of Th2 cell to elevate obviously.
