Objective: To investigate the effects of polycystin-1 N-terminal fragment(PC-1NF) fusion protein on proliferation and apoptosis of rat mesangial cell(RMC). Methods: Brdu-ELISA, Annexin V/PI binding, Real-time fluorescent quantitative RT-PCR, FACS, Electron microscope, Immunocytochemistry and Western blot analysis were applied in present study. Results: PC-1NF fusion protein may(1)regulate the cell cycle by modulating the PKCa signal pathway, concurrently inhibiting Cyclin D1 expression and augmenting P21Waf1 expression, subsequently blocking cell passing through G1-S check point, then inhibit the proliferation and production of extracellular matrix(ECM) of RMC. (2) induce apoptosis of RMC through increasing the ratio of Bax to Bcl-2 gene expression. (3) induce ECM degradation through increasing the ratio of MMP2 to TIMP1 gene expression. Conclusion: PC-1NF fusion protein may inhibit the proliferation, induce apoptosis and decrease ECM production of RMC.
