Effect Of Yishenhuoxue Decoction On Proliferation And Extracellular Matrix Accumulation In Rat Mesangial Cells

Objective: Mesangial cell proliferation and extracellular matrix accumulation are the major pathological characteristics of different kinds of glumerular diseases. Inhibition of cell proliferation and extracellular matrix deposition has an important role in preventing the occurrence and development of glomerulosclerosis.In our previous study,we have confirmed that Yishen Huoxue decoction improved the renal function and delayed glomerulosclerosis in rats, which is related to the inhibitory effects on the over-expression of TGF- β1,CTGF,CTGF,PCNA and FN and the alleviation of the accumulation of extracellular matrix. In this study,we shall observe the effects of Yishen Huoxue decoction on proliferation ,secretion of FN and COL IV ,expression of α -SMA and p-p38MAPK in rat mesangial cells,in order to investigate the mechanisms of Yishen Huoxue decoction.Methods:1.Preparation of pharmacologic serum: Twenty male Wistar rats, divided into normal control group and traditional Chinese drug group. Adopting the method of three-day intragastric administration, draw blood from the heart after the last intragastric administration. Then pharmacologic serum was prepared.2.Cell culture and groups: Rat mesangial cells were cultured with 1640 and were divided into three groups: normal control group,IL-1 β stimulation group and pharmacologic serum group.3.Detection of cell proliferation and cell cycle: After stimulation with 10% IL-1 β ,the proliferative activity was measured by tetrazolium salt colorimetry assay(MTT).
