OBJECTIVE To study umbilical cord blood (UCB) mononuclear cells(MNCs) differented into hepatic stem/progenitor cells in vitro and biological activity and transplantation of UCB-drived hepatic stem/progenitor cells in vivo. METHODS 1. UCB MNCs were isolated from UCB and cultured in the basic medium supplementedwith various combinations of growth /differentiation factors(FGF-l, FGF-2, LIF, SCF, HGF, and OSM). They differented into hepatic stem/progenitor cells, cell morphological change was observed, hALBmRNA was examinated using RT-PCR, the expressions of ALB, CK18,CK19 were detected by immunohistochemistry. 2. The liver-injured SCID mice were engrafted by UCB-derived hepatic stem/progenitor cells. In various stages after cell transplantation, hCD45 in their peripheral blood was analised by flow cytometry, the expression of hALBmRNA and hALB protein were detected by PT-PCR and Western-blot respectively, and the expression of hALB in the liver were examinated by immumohistochemistry. RESULTS 1. UCB mononuclear cells can differentiate into hepatic stem/progenitor cells in vitro and the positive expression of CK19,ALB by immunohistochemistry and hALBmRNA by RT-PCR were observed. 2. The liver was showed that its volume slightly enlarged, the surface of the liver had milium-like protuberance, sesamoid size. hCD45 was detected in peripheral blood by the flow cytometry. hALBmRNA and hALB were detected by RT-PCR and Westerm-blot. The positive expression of hALB were detected by immunohistochemistry.
