The new technique of proteome-protein antibody chip was used to detect the proteome expression spectum dynamically in the tumor tissue and serum of the patientswith epithelial ovarian cancer (EOC).The objectives of this study is to explore theexpression of differential proteins; to analyze the biological functions of abnormal expressed proteins; and to search the relationships between the abnormal proteome expression and the changes of immune state, female hormone and serum tumor marker of the patients with EOC. We try to reveal same relative factors to the occurrence and development of EOC; try to find biological molecules related or specialized to EOC; and try to supply new molecular markers for early diagnosis, evaluation of therapy and prognosis for the patients with EOC.This reseach includes following five parts.Part One The expression of differential proteins in the tissues of EOC by using protein antibody chipsObjective: To explore the differentially expressed proteins between normal ovaries and ovarian cancer tissues by using the protein chips (containing 512 cell function proteins). Methods: Tissues of 11 EOC and 11 matched normal ovaries were labeled with cy3 and cy5 fluorescent dyes and then were hybridized with 512 monoclonal protein antibody chips. The internally normalized ratio(INR)was calculated according to the intensity of fluorescence of each protein spots. The value of INR > 2.0 or < 0.7 was considered as the cut-value to filtrate the differentially expressed proteins between tissues of EOC and normal ovaries.
