Background and ObjectiveOLT (orthotopic liver transplantation) is the most effective method to treat acute liver failure, end-stage and metabolic liver diseases at present, but there are still some difficulties with widely clinical application because of insufficient liver donor and other reasons. HCT (hepatocyte transplantation) has some advantages including low cost, easily operation, reproductivity, meanwhile transplant cell can be cultured, proliferated, frozen stored in vitro, and genetically modified comparing with OLT. It has been found that HCT can correct the congenital liver metabolic defect, aid liver support, and replace damaged liver tissue. Along with the continuing development in theory and technology, HCT has shown the potential to substitute OLT for the treatment of liver diseases, but the main problems to limit HCT development are shortness of liver cell, poor ability of proliferation and immunity rejection after transplant.BMSC (bone marrow stem cell) can be horizontally differentiated into liver precursor cells and liver cells, and they can be easily acquired, cultured, generated and extended in vitro, and have strong ability to proliferate; they can be directly acquired from patients and thus the immunorejection can be avoided. The great breakthrough is expected in the treatment of liver diseases with BMSC instead of mature liver cells.BMSC can restore the liver function through two ways: one is direct BMSC transplant, then be differentiated into liver cell and proliferate, thus restore the liver; the other way: BMSC are derivated to directively differentiate into liver cells or liver precursor cells, generate and proliferate in vitro, then be transplanted.
