As a very complicated biochemistry process, the motablism of cells mainly involve in the dynamic changes and expression of various proteins, the activity status, the interaction between proteins, etc. With the ending of Human Genome Project, the major concern in life scientific research inevitably changes direction from genome to proteome. The newly emerging proteomics was developed on the basis of two-dimensional electrophoresis, by which the cellular protein mixture was separated and displayed in the polyacrylamide gel according to their different isoelectric points and molecular weights, images was subjected to laser scanner and computerized image analyzer, differentially expressed polypeptide spots were treated with physical or biochemical approaches (mainly the spectrometry technique for biomacromolecule), data obtained was subjected to searching of protein database(bioinformatics), so that the unknown proteins were identified. The composition, quantity of expression, modification and dynamic changes caused by various extrinsic factors of proteins in certain tissue or cell type can be displayed by way of proteomics. Proteomics provide a powerful tool for displaying, separating and comparing the pan-protein mixture of certain tissue or cell types, and find widely application in many researching fields such as tumor pathology, cardiovascular disease and drug development.Establishing the database of 2 dimensional polyacrylamide gel electrophoresis of certain tissue type paves the foundation for proteomics study. Comparative analysis of the proteome between normal and cancerous tissues will find the differential expression proteins, by extensive investigation, some protein markers might be picked out for the purpose of early diagnosis, prognosis evaluation, or served as targets for immunotherapy and medicinal therapy. At present, several large-scale proteome research programmes have been initiated including carcinoma of breast, carcinoma of colon and carcinoma of bladder, intending to establisth 2D-PAGE database respectively.
