Alzheimer's (AD) is the most common cause of senile dementia affecting many aged people worldwide. With increase of aged people this kind of diseases inevitably will rise rapidly in the future. It was estimated that there will be about 6 million people affected with dementia among people from 75 to 84 years old in the near 50 years. This number is about 12 million for the people over 85. It is clear that discovery of the drugs with therapeutic effect of dementia is urgent.Accumulation of β-amyloid peptide (Aβ or βAP or A-beta) in the brain is a defining feature of AD. More evidences indicate that Aβ can be neurotoxic in vivo and in vitro, and suggest that this protein may be actively involved in the neuronal degeneration that occurs in AD. Using Aβ as a toxic insult is an effective way in screening drugs with therapeutic effect of AD. Salvianolic acid-B (SalB) is one of the water-soluble components isolated from the traditional Chinese drug - Radix Salvia Miltiorhizae. Our previous studies demonstrated that SalB possesses several bio-activities, including alleviate ischemic damage in rats and neural cells. The present study was designed to investigate the effect of SalB on Ap induced neural cell damage.1. PROTECTIVE EFFECT OF SALB ON Ap INDUCED NEURAL CELL DAMAGE.Aged Ap(25-35)10μmol/l inhibited PC12 cells MTT reduction. SalB 10 nmol/1 -lμmol/ alleviated the toxicity induced by AP(25-35). After 30 h aging at 25℃, Aβ (1-40) 10μmol/L significantly inhibited the PC12 cells MTT reduction. The inhibitory effect became more apparent with an increase in the aging time, and this toxic effect was significantly reduced when the protein was aged in the presence of SalB. PC12 cells treated with AP(l-40) exhibited degraded DNA content characteristic of apoptosis cells (1.53% vs 19.9%).
