Part I. The Mitogen-activated Protein Kinase and Mast cells Involved in Cardiac Collagen Remodeling on the Basis of Morphology in the SHRsp.Background:1. A growing of evidences of molecular and cellular biology on hypertension indicated that cardiac collagen remodeling in association with fibroblast proliferating developed in cardiac hypertrophy. Collagen matrix may regulate myocyte behavour through acting on the mechoceptor integrin. Recently, A serine/therenine protein kinase refer to as mitogen-activated protein kinase(MAPK)with dual-specificity kinase are ubiquitously located in the cytosol in mammalian cells which play a central role in mitogenic signaling pathways stimulated by growth-promoting factors acting on a variety of cell surface receptor. In vitro study showed that MAPK activation closely related to the proliferation of myocardial fibroblast. So, activation of myocardial MAPK is purposed to play a pivotal role in cardiac collagen remodeling. To prove the hypothesis, the study are designed to investigate myocardial MAPK activity and its distribution combined with general morphology and ultrastructure of myocardium as well as collagen gene expressing in the hypertrophied heart and to clarify whether MAPK involved in the process of cardiac collagen remodeling in the SHRsp. 2.
