Objective: The research was to investigate the effects of YiqiHuoxue recipe on left ventricular remodeling, blood pressure and changes of rennin-angiotensin-aldosterone system(RAAS) activity in spontaneously hypertensive rats(SHRs) to further explain the mechanism of YiqiHuoxue recipe on preventing and treating left ventricular remodeling.Methods: 14-week-old SHRs were assigned randomly into 5 groups.-SHR-C(untreated), SHR-X(treated with nifedipine), SHR-Y1(treated with high dose of YiqiHuoxue recipe), SHR-Y2(treated with moderate dose of YiqiHuoxue recipe), SHR-Y3(treated with low dose of YiqiHuoxue recipe), each group consisting of 8 rats. After 8-week-treatments, we measured systolic blood pressure(SBP), heart weight index(HWI) and left ventricular mass index(LVMI); Sections of heart tissue was stained with HE method and VanGieson method. Cardiac tissue collagen - â…  and â…¢ contents were shown by labeled strept-avidin-biotin complex method(SABC). The changes of structure and shape in the myocardial cells and volume fraction in collagen were determined by automatically quantitative morphometry; We detected the concentration of aldosterone(ALD) and angiotensin â…¡(Angâ…¡) of plasma and myocardium with radioimmunoassay.Results: â‘ SBP was decreased in every treatment group. There were significant differences between pre- treatment and post-treatment of all groups(P<0.05). There were no significant differences among the treatment groups. HWI and LVMI in the SHR-Y1 group, the SHR-X group and SHR-Y2 group were lower than the SHR-C group (P<0.05). LVMI in the SHR-Y1 group was lower than the SHR-Y3 group(P<0.05). There was no apparent statistical significance between the other groups. â‘¡All collagen contents in the SHR-Y1 group were lower than the SHR-C group(P<0.05). Collagen-I contents with the SHR-X group and SHR-Y2 group were lower than the SHR-C group(P<0.05). All collagen contents with the SHR-Y1 group were lowerthan ones in the SHR-C group and the SHR-C group (P<0. 05). ?The morphological observation showed the collagens of myocardial and blood vessel were decreased in every treatment group. Both YiqiHuoxue recipe and nifedipine could restrain the synthesize of collage. ?The concentration of ALD and Angll of plasma and myocardium with the SHR-Y1 group and the SHR-Y2 group was either lower than the SHR-C group (P<0. 05). The concentration of ALD and Angll of myocardium in the SHR-X group was lower than the SHR-C group (P<0. 05).The concentration of Angll of plasma and myocardium and ALD of myocardium in the SHR-Y1 group was lower than the SHR-C group(P<0. 05). The concentration of Angll of plasma and myocardium with the SHR-Y1 group was lower than the SHR-Y3 group(i><0. 05).Conclusions: ?Both YiqiHuoxue recipe and nifedipine could inhibit LVH while them had no apparent statistical significance. ?YiqiHuoxue recipe could decrease myocardial f ibrosis, the effect of YiqiHuoxue recipe was better than that of nifedipine. ?Both decreasing blood pressure and restraining RAAS activity could be the mechanism of YiqiHuoxue recipe preventing left ventricular remodeling.
