Epithelial ovarian carcinoma represents a great clinical challenge in gynecologic oncology. Since most patients have metastasized when they are symptomatic. Using a multimode treatment including surgery and combination chemotherapy, five-year survival rates are just about 30%. The immunotherapy will be helpful to the patients. Tumor antigens play important roles in initiating immuno-response and acting as targets for immuno-attack. Cancer immunotherapy strategies based on antibody have shown great success in recent years. The excellent example is Herceptin, a humanized antibody specific to P185erbB2, which shows good curative effects in metastatic breast cancer. As P185erbB2 overexpress also in ovarian carcinoma and concern with poor prognosis and drug resistance, we then prepared humanized McAb against P185erbB2 and high-level expressed in CHO cells for adjuvant therapy of ovarian carcinoma. To overcome the deficiency of immunotherapy aim at single tumor antigen, that caused by heterogenic trend of tumor cells and preferential outgrowth of antigen-negative clones under the selection pressure of single antigen vaccine or single antibody, we tried to identify novel ovarian carcinoma antigens to improve immunotherapy efficiency for future by exploiting the pool of ovarian carcinoma antigens.Preparation and identification of McAb specific to human P185erbB2 then validity test in adjuvant therapy of ovarian carcinoma in nude mice.BABL/c mice were immunized with SKBR3 cells over expressing P185erbB2.
