Trichinella spiralis is the genus of parasitic roundworms of the phylumNematoda. As a zoonotic parasite, it can infect various mammals which alsoincluding human. Numerous studies have shown that Trichinella spiralis infectionhas anti-tumor effect. In our lab, it has been identified that Trichinella spiralisrelated antigens have inhibitory effects on both lung cancer cell line A549andbreast cancer cell line MCF-7. Liver cancer is the leading cause of death inworldwide. Although the liver plays the major role in drug metabolism, hepatictissue also easily get the drug resistant. Thus, there is an important need for developnew therapeutic strategy for treating liver caner. Monoclonal antibody drugs are arelatively new innovation in cancer treatment. In this study, we aimed toimmunoscreen the related antigen between Trichinella spiralis and H7402cell andprepare the monoclonal antibody to test if the monoclonal antibodies have ananti-tumor effect.ELISA and Western blotting were used to explore whether there are associatedproteins between Trichinella spiralis and human hepatoma cell line H7402. Thenimmunoscreening technic was used to get associated genes between Trichinellaspiralis and H7402cells. After that, the genes were cloned by PCR and expressed inE. coli. In the end, we got8related antigen genes between Trichinella spiralis andH7402by immunoscreening, they are: Tsp02534, Tsp09956, Tsp07163,Tsp10907, Tsp10072, Tsp00188, Tsp06120, Tsp10907. Later on Tsp10907(CK-1) was picked out. CK-1protein’s monoclonal antibody was prepared, and itsanti-tumor was tested in both vivo and vitro. In vitro, the MTT assay showed thatthe proliferation of H7402was significantly inhibited by monoclonal antibody,when the concentration of monoclonal antibody was250μg/mL, the cell inhibitionrate is39.23%. In vivo, the anti-tumor effect of monoclonal antibody was tested in H7402tumor burden balb/c nude mice. The highest inhibition rate is39.23%whenwe inject mice with antibody concentration of500ug/mL. We also checked theapotosis protein expression in the mice tumor after the treatment byimmunohistochemical. Results showed that the expression of P53, VEGF, Bcl-2intreated group is lower than the control group.This research lay the foundation for the studies of anti-tumor effect ofTrichinella spirallis and provides a scientific basis for the development ofanti-tumor biological agents.
