[Part I II] Objective To research the protective effects of drugs H Y and D on cold preservation injury of rat livers and synergistic action of three kinds of drugs by combined use. Method Isolated perfused rat liver model was used.We observed the preservation effects of Lactate Ringer's (LR) solution added various dosages of drugs H Y and D after 12 hours cold storage of rat livers by the four parameters including measurement of adenine nucleotide,dihydroxybenzoic acid (DHBA) and bile production,and assay of AST in the effluent .Results The contents of ATP TAN AEC and bile production in the groups of LR solution with the addition of drugs H, Y and D were significantly higher than those of control group (P<0.05),while the changes of the contents of DHBA and AST were just on the contrary .Within certain range of the dosage of the drugs H Y and D ,the content of ATP and the amount of bile production were in the positive ratio of the dosage in the LR solution.Conclusions The significant protection of drugs H Y and D on liver preservation revealed in this research may act by a mechanism which correlated to the effect of improving the energy metabolism of cold storage liver by drug H,resisting free radical by drug Y and both actions by drug D.Three kinds of drugs H Y and D by combined use in presevation solution show obvious synergistic action.[Part III] Objective To evaluate the cold preservation effect of rat livers with self-made HYD solution. Method We made the comparisons of the preservation effect among HYD solution,UW solution and LR solution by using isolated perfused rat liver model.Results All the parameters of hepatic biochemical functions and morphological observation in HYD Group and UW Group were better than in LR Group for 12 hours storage (P<0.05).The hydrolytic activities of H~+ -ATPase in hepatic mitochodria and bile production were slightly higher in HYD Group than in UW Group(P>0.05).Dry weight/wet weight rates of liver tissue in HYD Group were similar with those of control group and were markedly lower than those of UW Group .The values of pH in the...
