OBJECTS:Pantothenic calcium, is the component of CoA, which takes part in the metabolism of fat ,carbohydrate and protein in the body. In order to explore effects of pantothenic calcium on starved animals and provide the basis for developing survival ration, we studied the protective effect of pantothenic calcium on starved rats and mice and its probable mechanisms.METHODS:1.Protective effect of pantothenic calcium on starved rats and mice. The adult mice and rats used as fasting animal model were divided into four groups: starved control group(C group), low pantothenic calcium supplemented group(L group), middle pantothenic calcium supplemented group(M group), high pantothenic calcium supplemented group(H group). During the fast, they all received water ad libitum. We observed their survival time , body weigh chang and urine nitrogen excretion. In addition, we invetigated the effect of pantothenic calcium on brain function by mesuring the ability of learning and memory with Y maze in mice.2.Mechanisms for protective effect of pantothenic calcium on starved rats and mice. SD rats were divided into seven groups: nomal control group (NC, n=6), starved control groups(n=24, include three groups), supplemented groups(n=24, include three groups). We used optimum dosage from preivious experiment as supplemented dose, which were treated as before. Rats were killed at zero of fasting in NC group, others in starved control groups and supplemented groups were killed at four, seven, nine days of starvation~spectively and the concentration of serum ketone bodies and free fatty acids were detected. The level of MDA, lactate and OSH , the activities of GSH-Px, SOD, Na~,K~-ATPase,Ca2~-AXPase and LDH were examined in rat brain tissue, apoptosis was dectected in rat brain as well.RESULTS:l. ProtCctive effect ofpatthenic caIcium on swted tals and mice.PNc caIcium couId Prolong the survivaI time, and significanly in M grouP,in which Phase II was extrended mainly It was suggested tha pantothenic calciumcan sighficanIy prolong sUrVval time and phase II in the starved rats and mice. The abilihes of ledng and memory of ndce in M and H grOuPs was sighficantlyhighe than thOse in C grOuP by analysis of covariance at the third day of fasing. Itwas indicated pantothenic calPium have PrOteCive effect on StarVd mice btaln.2. Mechbosms for PrOtective effect ofpantothenic caicdri on thed rats and miceThe effects on serum KB, FFA, Ca2+ conton in trid rats: KB and FFAinereased raPidly following food dePrivation in all rats and then decreased. Peaklevels in the suPPlemented rats wee lower than thOse in C grouP, esPically FFA leveldeclined markdly When energy almost dePleted at nine days off asing, KB and FFAcontent in the SUPPlemented rats wee sighficanly higher than total fasting rats, andserum Ca2+ level had no difference. It was indicated pantothnic calcium can slowdown eneny dePleted and pantothenic acid mak the main role. The effects on anti-oxidative system in thed rat bndn: Lipid peroxidaionMDA colltCnt incleased markedly ther fasting, hOwevr MDA cODted in suPlementeStarVed rats rose no sighficanly comPared with normai rats at four and Stwe days offasting, and had markd dismpancy comPared with beed control grOuPs.Pantothenic calcium had no aPparellt effectS on the activities of SOD, GSH-Px, andsighficantiy enhanced GSH levels in the stwed rats bu Whch were lowesighficanly than tha before fasing. It was suggested tha Pantothenic calcium canprOtect against hannful liPid Peroxidation in rat btaln and some of the mechanisms beprobably due to eboing the level of GSH.The effects on ATPase,LDH activity and lactate levels in the StarVd rat bndn:The actiVity of Nat K+AThase, C a2 +ATP ase and LDH drOPPed at vari ous de gree s,and lactic acid rose uP after fasting. Pantothnic caIcium could mainndn the activityof Na, K+-N,Ca2+M' at certain levels tO thect btaln fimCton. The effects on aPOptosis in starVd rats bndn: ApoPtosis in rat brain incleasedsi...
