Part I Isolation,culture and label of neural stem cells from newborn guinea pigObjective The research of neural stem cells has become more and more important in many medical fields all over the world. Neural stem cells are an adult stem cells.They have the powerful plasticity and ability of renew.They can be used in the research of cell therapy and gene therapy.The objective of this experiment is to establish a simple and reliable method for isolating,culturing and labeling neural stem cells derived from guinea pig.Materials and Methods The brain tissues from newborn (24h) hippocampusof guinea pig were isolated. After proliferation in conditional culture medium including bFGF or fetal bovine serum, the cells was observed by Nestin,GFAP and NSE immune fluorescence staining .At last the neural stem cells were marked with fluorescent dye DAPI.Results 1. The cells in primary culture became suspend neruospheres in differentsizes.The borders of the neruospheres were clear and the reflection is satisfactory.
