Objective:Now obesity is a worldwide epidemic disease similar sharp increase in the prevalence of obesity have occurred worldwide. Overweight patients are at risk for developing a number of medical social and psychologic disabilities, especially coronary heart disease, stroke, hypertension, type 2 diabetes, hyperlipidemia, sleep apnea et al. There are many factors influencing the development of obesity. At the center is the balance of energy that people take and consume in daily life. Excess energy is stored in the white adipocyte, and the proliferation and differentiation of preadipocyte is the core of the development of mature adipocyte. Many cytokines and hormones take part in the regulation of the development of adipocyte. To find out these regulating mechanism helps us to understand the pathogenesis of obesity, and to find out new targets for weight reducing drugs. Insulin-like growth factor-I and growth hormone are the most important cytokine and hormone related to growth in vivo. And Ghrelin, a new polypeptide found in stomach, causes weight gain through neuropeptide Y. The aim of the this study is to study the effects of the three cytokines and hormones of the proliferation and differentiation of human preadipocyte and mice 3T3-L1 preadipocyte in serum-free medium.Methods:1. To set up the culture of mice 3T3-L1 preadipocyte in serum-free medium .And to establish the method of primary culture of human preadipocyte with collangenase...
