Objectives: In this study, we had planed to use RT-PCR technique to clone hIL-24 Gene from pBV220-hIL-24, and reconstructed that gene with prokaryotic vector pET-21a(+) as pET-21a(+)/hIL-24. The rhIL-24 protein expressed by pET-21a(+)/hIL-24 in BL21 was used to treat PBMC, CAM, A375 cells and A375 cells'melanoma transplanted to nude mouse. We examined the cytokine secretion profile of PBMC, angiogenesis-suppressing in CAM, growth-suppressing in A375 cells in vitro and A375 cells'melanoma transplanted to nude mouse in vivo after rhIL-24 treatment. That study paved the way for the clinical usage of rhIL-24.Methods: We cloned hIL-24 gene from prokaryotic vector pBV220-hIL-24 and reconstructed prokaryotic vector as pET-21a(+)/hIL-24, transfected them into BL21(E.coli.), by the induction of IPTG and gained high expression of rhIL-24, the expressed protein was extracted, purified and renatured as rhIL-24, checked the product with Western-blot. Treated the PBMC with rhIL-24, collected the cells'culturing medium supernatant, and checked the cytokine IL-6, IFN-γand TNF-αwith ELISA. rhIL-24 was used to treat A375 cells in vitro, MTT tests were used to evaluate the influence of rhIL-24 on the growth of A375 cells. The growth curve of A375 cells was depictured and the IC50 of that reagent was calculated. FCM and fluorescent staining were used to show the influence of rhIL-24 on A375 cells'cell cycle and apoptosis phenomena. The angiogenesis-suppressing phenomena were observed in CAM after rhIL-24 treatment.
