Melanoma is a serious malignant skin cancer that is still seeking a drug with good efficacy.Mongolian medicine is used for cancer treatment and tumor angiogenesis is considered a major sign of cancer development.We first injected melanoma cells into vascular transgenic zebrafish embryos and made a model of zebrafish juveniles with melanoma.Then the therapeutic effect and mechanism of 2,4-dihydroxy-6-methoxy-3-methylacetophenone(ECB)were discussed.The results showed that the ECB can induce the apoptosis of melanoma cells and inhibit the proliferation of melanoma cells in vitro.ECB was similar to inhibitor Vatalanib(PTK787)2HCL,which not only inhibited the number and length of subintestinal vein(SIV)in zebrafish embryos(p<0.05),but also inhibited the metastasis and distribution of melanoma in zebrafish juveniles(p<0.05).More importantly,unlike the vascular inhibitor PTK,EBC only inhibits abnormal growing blood vessels.It was found by gene level detection that ECB also inhibited the vegfa?vegfr2 and vegfr3 m RNA expression which regulates blood vessels and Up-regulation of caspase-3and p53 gene expression.The above results indicate that ECB has a certain anti-melanoma effect,which may be achieved by inhibiting vascular receptors and promoting apoptosis.
