The pathophysiology of acquired aplastic anemia can be defined as immunologically mediated decrease in the size of the stem cell compartment. Immunomosurppressive treatment is the approach to most patients not eligible for sibling donor BMT . Co-stimulatory signal is the key element to initiate T cell immune response or arise anergy to self-antigen.In this study,we observed the effect of co-stimulatory associated molecule CTLA-Ig,anti-CD40LmAb on the T cell activation and proliferation and hematopoiesis recovery Part â… Study of the regulation of the T cell anergy in aplastic anemia patients by CTLA-4Ig fusion protein or anti-CD40LmAb in vitroObjective : To investigate the inhibition of the abnormal bone marrow T lymphocyte activitation and proliferation by the CTLA-4Ig fusion protein/anti-CD40 ligand monoclonal antibody (4F1) in vitro in patients with aplastic anemia(AA). Material and methods:Bone marrow samples were collected from 25 AA patients and 10 patients with non-hematopoietic diseases.BMMNCs were prepared by ficoll gradient centrifugation technique.BMMNCs were resuspended in the medium containing RPMI-1640,2 mmol/L...
