Part One Research on biocompatibility of the valved bovine jugular vein conduit in vitroBackground: The valved bovine jugular vein conduit is a novel biomedical material for reconstruction of right ventricular outflow tract in cardiac surgery. There is no available material in China at present.Objective: To evaluate the biocompatibility of the valved bovine jugular vein conduit with different treated methods in vitro by using the hemolytic test and the cytotoxicity test.Methods:1. Blood compatibility in vitro: To evaluate blood compatibility, we observe the hemolytic rates, reticulocytes count and blood routine count by using blood direct contact with the valved bovine jugular vein treated with different methods in vitro.2. Cytotoxicity test: To evaluation the cytotoxicity of extracted liquid of the valved bovine jugular vein conduit on the proliferation rate of L-929 mouse fibroblasts, we use direct cell contact evaluation and MTT colorimetry method. The growth of cells was observed and the number was counted after 24, 48, 72 hours in direct cell contact evaluation test. Absorbance of all the tested tubes was measured by a spectrophotometer at 492 nm after 2, 4, 7 days respectively in MTT test.Results:1. The hemolytic rates of the valved bovine jugular vein conduit treated with glutaradehyde(GA), polyepoxy compound(PC), GA/octanol, dye-mediated photooxidation(DMP), and decellularization combined with dye-mediated photooxidation were -0.10%, 0.33%, 0.33%, 0.05%...
