Part I Nanobacteria culture, harvest and identificationObjective: To explore the culture, harvest of nanobacteria, its material consumption under culture, and its growth speed and identification.Methods: Desirable culture condition, harvest and antipollution method for nanobacteria were investigated. Changes of electrolyte and glucose were dynamically detected by auto clinical chemistry analyzer. Also the absorbance of nanabacteria was measured by Elisa to know its growth speed indirectly. Immunofluorescence was then conducted to identify the cultured nanabacteria by applying the cloning antibody of the nanabacteria. Finally, nanabacteria was observed with electron microscope.Results: The desirable culture condition of the nanobacteria were: using Cell culture method, 10% culture medium of FBS with nanobacteria added, then put into the incubator with the atmosphere of 5% CO2 and 95% air at 37℃. A range of measures were taken to prevent the nanobacteria from being polluted during its culture.
