PARTⅠEXPRESSION OF DOUBLECORTIN DURING THE NEURONALGENESIS OF ADULT RATSObjective:To investigate the expression of the microtubule-associated protein, Doublecortin (DCX) of neuronal precursors during the neurogenesis of adult rat, and provide the theory and experiment basis for the study of neuronal proliferation and migration.Methods:The adult rat brains were cut into 20-μm coronal and sagittal sections by using a freezing microtome. Immunohistochemistry and DAB coloration were applied to observe areas and morphology of DCX-expressing cells under light microscope.Results : The DCX-expressing cells distribute mainly in five neurogenic areas: subventricular zone, subgranular zone, rostral migratory stream, olfactory bulb and around the corpus callosum. However, only very low levels of DCX expression in the nonneurogenic regions such as cortex. The DCX-positive cells have the fusiform somata with a single leading process.Conclusion : The DCX-positive cells have the characteristic morphological features of neuronal precursors. DCX could be used as a marker of neuronal precursors for the study of neuronal proliferation and migration.
