Objective To make minority herbs combine with modern technology, studying the effects of Osteoking (herbs of the Yi minority in Yunnan province) in the treatment of femoral head necrosis relevant to steroid.Methods 1. According to documentary reports and many pre-experiment results, we made rabbit model of SANFH with the method of twice intravenous injection with low dosage LPS (10μg/kg body weight, at a time interval of 24 hours.) and 3 times intramuscular injection with MPS(20mg/kg body weight, at a time interval of 24 hours.); detected the quantity of the blood serous CHOL, TG and HDL of this model; observed the changes of histology and ultra-minimal structure of femoral head through HE, TEM and SEM; at the same time, compared it with the model only using PS intramuscular injection. 2. After the treatment with Osteoking, the changes in blood fat and the structure under HE and TEM were compared between the two models. 3. Then make further investigation using the model treated by LPS and MPS: At the 4th week after the model built, 4 rabbits were chosen randomly and proved to have femoral head necrosis examined by MRI and histology, then rabbits were divided into 4 groups randomly: group A (blank control group), group B (normal rabbit fed with Osteoking), group C (model group without feeding Osteoking), group D (model group fed with Osteoking). There are 24 rabbits in each group. Once every 2 days, irrigate the stomach in group B and D with Osteoking, while A and C with normal saline. At the 4th week, selected 4 rabbits after the blood fat had been detected. At the 4th, 8th and 12th week, took 8 rabbits in each group and put them to death after the blood fat had been detected. Then take out both femoral heads, cut coronal section to observe roughly and keep the bone for HE, TEM and SEM observation and made comparison with the results. 4. At the same time, make immunohistochemistry, in situ hybridization and extract total RNA for timely fluorescence ration PCR. In this process, we only use 4 hours to get total RNA from femoral head.
