Objective: 1 , To build skillfully the model of orthotopic liver transplantation(OLT) in Wistar rats without rejection, then to build the model of OLT from DA to Lewis rats with acute rejection. 2, To study the function of HO-1 in the model of OLT when it is induced or inhabited by inductor CoPP or suppressor ZnPP respectively, and its effect to pro-inflammatory molecules ( IFN-γ ,IL-12 ) and cytoprotective molecules ( IL-4 , TGF-β1) .through these models to observe different affections to the function of liver cells, infiltrate of inflammatory cells ,degrees of rejection , changes of BCL-2 and Adhesion molecules in rats according to the different expressions of the HO-1.By all these experiments, we want to elucidate the anti-inflammatory and cytoprotective functions of HO-1 in the OLT in rats. Methods: 1 , OLT without rejection in rats was performed by the "improved tow cuffs" method. 2, To build the OLT with acute rejection in rats by the same method, then rats were divided into three groups: control group(A), ZnPP group(B) and CoPP group(C); 3 or 7 days after the operation, recipient rats were killed, blood and samples were collected.
