Objective: To observe the healing of osteoporotic fracture and to study effects of L-arginine on the healing of fracture in ovariectomized rats. Methods: 1. Osteoblasts were isolated from rats' cranium by enzymatic digestion and were verified to be typical osteoblasts by this method. Then the cultured osteoblasts were interfering with different concentration of L-arginine and PTH. The OD values in every group were measured by MTT assay; the variability in protein OPG was evaluated by western-blot test; the influence of eNOS and iNOS in osteoblasts were observed by immunohistochemisty technology.2. Osteoclasts were cultured from neonatal rats' long bones by marrow induction method and were verified to be typical osteoclasts by this method. Then the cultured osteoclasts were interfering with different concentration of L-arginine. The change of values and shapes of osteoclasts were observed with contrast phase microscope everyday; after osteoclasts were stained in TRAP, it is to count cells in TRAP (+) and to record the change of these cells; the area and number of resorption pits were observed with scanning electric microscopy at the same time.3.
