Bone morphogenetic protein-2 (BMP-2) plays a decisive role during bone regeneration and repair as well as in various stages of embryonic development. The crude and partially purified bone -derived bone morphogenetic protein combin- ed with various carriers has been successfully applied clinically to treat bone defect and bone nonunion and demonstrates bright prospects. But it is difficult to purified BMP from bone matrix. Since the 1980s, the recombinant hBMP-2 has been expressed in COS, CHO and insect cells as a functional active form, but the low output and high cost restricts its application. Zhao ming, Liu xinping et al expressed modified C-terminal peptides of hBMP-2 in E.coli independently, and after purification and renaturation BMPS was prepared as a functional active form. The hBMP-2 expressed in E.coli makes up the deficiencies mentioned above. In this paper, the inductive mechanism and bone forming effects of rhBMP-2 are studied to provide a theoretical basis for its application.(1) The inductive effects of rhBMP-2 in vitro and in vivo.
