Anti-hepatoma McAb and the β-glucuronidase cDNA will provide useful tools to study hepatoma targeted diagnosis and gene therapy. In this paper, a serial experiments , 99mTc labeled McAb HAb18 for radioimmunoimaging (RII) and β-glucuronidase cDNA used for hepatoma "suicide gene" ,were done . The whole studies were summarized as follows:1 99mTc labeled anti-hepatoma McAb HAb18 fragment for RII in nude mice bearing xenografted human hepatoma . Firstly, the anti-hepatoma McAb HAb18 was digested by active papain at different pH condition. At pH5.5, 37℃ for 18h, F(ab')2 fragment was obtained; and Fab fragment was at pH8.0 37℃ for 1h , the production was purified by DEAE-FPLC。 The fragment purity was more than 90%. The yield was 40% to 60%. Secondly, In order to increase the sulfhydryol group , McAb Fragments were modified with 2-iminotholane and labeled with 99mTc by transchelation from 99mTc-GH .Finally, the 99mTc-IT-F(ab')2 and 99mTc-IT-Fab were injected into the nude mice for RII.
