Thrombosis is one of the pathogenys which responsible for very high mortality resulted from many cardiovascular diseases. It is urgent to develop new drugs for anti-thrombotic treatment. Dihydropyridines(DHPs) are used therapeutically in treating hypertension, acute myocardial infarction and various other ischemic disease states, and some compounds of DHPs have been reported to have the power to inhibit platelet activation. Till now, few studies about effects of DHPs on in vivo. thrombosis models has been reported yet. The purpose of this experiment was to investigate the anti-thrombosis effects and mechanisms of Methyl pentyl 1,4-dihydro-2,6-dimethyl-4-(3-nitrophenyl)-3,5-pyridinedicarboxylate(MN9202), a novel DHPs calcium antagonist, on three experimental thrombosis models induced by carrageenan or platelet stimulants.1. Tail thrombosis model Duplicated by carrageenanAdult male Sprague-Dawley rats, LACA mice were given a single sc. carrageenan (rat:5mg/kg, mouse: 20mg/kg) to induced the tail thrombosis, 24h and 72h later, extravasated blood, black appeared in tail of the rats at 90% and mice at 100% respectively. Pathological observing indicated: Mixture thrombus composing of fibrin , RBC , platelet and WBC existed in tail arterial vascular, Endothelial cell was injured.
