To developing effective therapy for rheumatoid arthritis (RA), suppression of adjuvant arthritis (AA) by oral administration of type II collagen (CII) was studied in term of the therapeutic efficacy and its mechanisms in the present experiment.CII was extracted from bovine cartilage by pepsin digestion, was purified by DEAE-cellulose (DE-52) column chromatography, and was identified by SDS-PAGE analysis and thin-layer scanning of SDS-PAGE gel. The purity of DEAE-purified C II was > 95%.Rat adjuvant arthritis is induced by intradermal injection of complete Freund's adjuvant (CFA), and used as a model of RA in the present experiment. It was demonstrated that Adjuvant arthritis not only could be suppressed by oral administration CII but also suppressed by adoptive transfer of splenic cells (SC) and mesenteric lymph nodes lymphocytes (LC) from CII orally tolerized rats in a dose-dependent manner. The disease onset was delayed and the severity was reduced.
