Objective:Observe the effect of Chinese herbs treat Adjuvant Arthritis which is most frequently used in Rheumatoid Arthritis including diaphoretics, wind-Expelling herbs, blood activating drugs and compound mixture. To study its synergism and compatibility rule and the Chinese herb which play dominant role in mixtures, explore the mechanism of Chinese herbs treating Rheumatoid Arthritis.Methods:1. Healthy male 24 wistar rats were selected and divided into 2 groups by random: model group and normal group.Every rats of model group were injected 0.1ml complete Freund's adjuvant with BCG 10mg/ml into the rat left hinderpaw to induce adjuvant arthritis by intradermic injection. Normal group were injected 0.1ml physiological saline into the same position. The pathological changes of adjuvant arthritis were investigated over a period of 22 days after induction of this model. The swelling rate of both hinder paw every rats were measured on alternate day. the rats were killed on 22th day, and each ankle of right hinder leg was made into pathological slice to confirmed the Adjuvant Arthritis had been made2. Healthy male 84 wistar rats were selected and divided into 7 groups by random: model group, dexamethasone group, wind-Expelling group, diaphoretics group, Blood activating group, compound group and normal group. Adjuvant Arthritis was made except normal group. Each group gives the different drug mixture:the rat drug dose is calculated According to formula and rat weight. The pathological changes of different groups were investigated over a period of 22 days after intradermic injection. The swelling rate of both hinder paw every rats were measured on alternate day to observe therapy effect in defferent groups. The blood samples were collected from the heart under light ether anesthesia after had fed for 22 days by gavage, then the serum NO/MDA level and SOD activity were measured. Three rats of each group were selected to cut the right ankle, and the ankles were made into pathological slice to observe the change. The TNF-α/IL-1β/PGE2 level of right paw and joint were measured in other nine of each group. Every rat was weighted spleen and thymus which cut down from the rats for its organ index.Result:1. The left hinderpaw of model group swelled obviously in the next day, and reached the peak at 3th day, the swelling got better gradually thereafter, and right hinder-paw appeared red swelling at 11th day and reached the peak at 15th day. The swell-ing got better gradually thereafter until the experiment was finished. A small number of rats appeared red swelling on the ears and tail. The synoviocyte hyperplasia and a large number of inflammatory cells with pannus formation were found in the synovium of model group rats through pathological examination. It showed Adjuvant Arthritis was made successfully.2. compared with the model group, the paw swelling of compound group/wind-Expelling group and dexamethasone group had fed by gavage for 22 days relieved evidently. And there was no statistical difference between compound group and wind-Expelling group.the diaphoretics group and blood activating group relieved to a certain extent. Compared with compound group, the therapy effect of blood activitating group was inferior to compound group; also the therapy effect of blood activitating group was evidently inferior to compound group3. The TNF-α/IL-1β/PGE2 level of right paw and joint in compound group/wind-Expelling group was lower than model group evidently, PGE2 level of right paw and joint in diaphoretics group was lower than model group evidently, and no evidently statistical difference was found between blood activating group and model group. The TNF-α/IL-1β/PGE2 level of right paw and joint in diaphoretics group and blood activating group were higher than compound group4. The NO/ MDA level of serum in compound group/ wind- Expelling group/ dexamethasone group and blood activating group were lower than model group. The SOD activity was higher than model group. No difference was found between model group and blood activating group.5. Compared model group with normal group, spleen and thymus index of model group was higher than normal group, dexamethasone group decreased evidently, and other groups had no difference with model group. 6. Pathological slices in different group showed:few rats were found which have synoviocyte hyperplasia and inflammatory cells with pannus formation in the compound group and wind- Expelling group, While there is a few pathological change in diaphoretics group, blood activating group.Conclusion:1. Adjuvant arthritis is a suitable model animal which is used for studying rheumatoid arthritis.2. Compound mixtures / wind- Expelling mixtures /Blood activating mixtures and diaphoretics have therapy effect on the swelling paw of AA rats, the compound mixtures have evidently effect. No evidently difference was found between compound group and wind-Expelling group, so wind-Expelling mixtures may play a major therapeutic effect in the compound mixtures.3. Decreasing TNF-α/IL-1β/PGE2 level in paw and joint is the action mechanism of compound mixtures and wind-Expelling mixtures, and both mixtures can decrease NO and MDA level of serum, increase SOD activity of serum. Blood activating mixtures also can decrease NO and MDA level of serum, increase SOD activity of serum. Diaphoretics can only decrease PGE2 level in paw and joints. All of above show:it is by many different ways that Chinese medicine treat rheumatoid arthritis...
