In the present study, the light and electron microscopic localization of lectin receptors on human gastric cancer cells and epithelial cells in chronic gastritis were studied by means of immunogold probes prepared by our own. The simultaneous ultrastructural localization, of the corresponding antigens of antigastric cancer monoclonal antibodies and lectin receptors on human gastric cancer cells were also performed.The satisfactory results of using streptavidin-gold and goat antimouse IgG-gold in this study indicated that the immunogold probes possessed many advantages, such as higher specificity and sensitivity, economy and simplicity. They could be used for light or electron microscopy and single or double labeling, blent immunocytochemistry with ultrastructural observation. Indeed, they were good immunocytochemical methods with glorious future.The light microscopic localization of 5 lectin receptors showed that there were some differences in lectin receptors expression between cells from gastric cancer and chronic gastritis as well as between different histopathologic types of gastric cancer, that the positivity rate of PHA in gastric cancer was 97.56% , that the strong positivity rate in gastric cancer was higher than that in chronic gastritis (P<0.05), and that the strong positivity rates of WGA in mucous type of cancer, of DBA in poorly differentiated cancer were higher than that in other types of gastric cancer. The results suggested that comparing these differences might have help to the diagnosis of gastric cancer.The electron microscopic localization of 3 lectin receptors showed that positive gold particles were mainly localized on membrane structure of cells, that expression of lectin receptors in gastric cancer cells was enhanced obviously. The results suggested that cell membrane structure was major place where glycoprotein was synthesized, transported, and secreted; that carbohydrate metabolism and mucous secretion in gastric cancer were vigorous.
