Objective: Using an animal model with nerve cutting dorsal root of chord spinals, which had been made large gap in the continuity of the sciatic nerve and the gap was repaired by acellular nerve allograft made through chemical extraction, to study the characteristics of the chemotaxis during the nerve regeneration. Methods: 30 rabbits were randomly divided into the experimental group (16 rabbits) and the blank group (14 rabbits). Observe the change of morphology and histopathology after the animal model was established. After 3 weeks, the 2 groups were both cut of a small gap of right sciatic nerve (contain sciatic nerve trunk and crotch of tibia nerve and fibular nerve) to make up the neurologic of defect sciatic nerve. At the same time, to sew up sciatic nerve involucrum with acellular nerve allograft made through chemical extraction conduct and actions Y shape stand. 3 months later, contrast the differences between the 2 groups of the posterior limb, nerve conduction velocity (NCV), and cortex sensory evoked potential (CSEP) and histopathology. Results: The experimental group gradually appeared ulcer of the posterior limb foot and had obviously change of histopathology. It was showed that nerve tissue had part of neuron edema, loss and disintegration of myelin sheath. After 3 months ① The controlled group motor function of posterior limb had recovered gradually, and the foot ulcer had also recovered to normal level; The experimental group motor function of posterior limb had recovered gradually too, however, the foot ulcer was still apparent. ②Electro physiology detection showed that the controlled group and the experimental group could be measured the nerve conduction velocity on the section of neural transplantation, and there were no significant differences in the 2 groups(p>0.05), especially the inferior of uninjured side; The piece of neural transplantation could be measured cortex sensory evoked potential in the controlled group and the experimental group was not. ③After 3 months of neural transplantation , there were abundant new nerve fiber in both controlled group and the experimental group; And there was a great quantity modulated nerve fibers to be generated in distal end. The density of neuro-fibra reproductionin tibia nerve of controlled group is 1635.66±187.42piece / mm2,which is super to experimental group, 1125.00±144.12piece / mm2 was observed through the electron microscope . The content of aesthema rami average regenerate neuro-fibra was 458 pieces in controlled group, and 25 pieces in experiment group, differences of statistics is significant (p<0.05). Conclusion: The experiment indicated that the animal model of rabbit dorsal nerve root of lumbar could be successfully established with cutting the dorsal nerve root of spinal cord and homogeneity same sciatic nerve transplantation, which were both tested chemotaxis through the electro physiology and histopathology detection during periphery nerve regeneration.
