OBJECTIVE: Gliomas are the most common primary brain tumors. Although currently available combined therapy including operation , radiotherapy , chemotherapy are used, the prognosis of patients with malignant gliomas is still very poor. The activation of cellular proto-oncogene and the deactivation of cellular antigene play very important role in the process of tumor genesis. As the knowledge of tumor biology and molecular genetics increased, people keep exploring various kinds of new treatment methods to suppress the glioma cell proliferation for improving the result of treatment. It has been verified that alteration of multiple genes is involved in the molecular pathogenesis of tumor development. So the gliomas is the same as the tumor in other sites of the body, it's a disease involving multiple genetic changes. As a recently detected tumor suppressor gene, PTEN is the first one who deserves the phosphotase activation. Our research aims to discuss the gene and protein expression of PTEN in human gliomas, to find out the dependablity between the PTEN expression with the tumor malignancy grading; also, we combine with the results of cellular proliferation and apoptosis of human gliomas, in order to analyze influence of PTEN gene expression on the apoptosis of human gliomas and the relationship between them. And we can make better understanding of the role of PTEN in the process of tumor formation and development to provide theory...
